Researchers subjected fibroblast matrices to various strains and employed a scratch wound strain model to test the ability of OMT to impact wound healing.

In a typical scratch wound healing assay, a “wound gap” in a cell monolayer is created by scratching, and the “healing” of this gap by cell migration and growth towards the center of the gap is monitored and often quantitated. Factors that alter the motility and/or growth of the cells can lead to increased or decreased rate of “healing” of the gap (Lampugnani, 1999).

Human fibroblasts were seeded onto 6-well collagen-I bioflex plates, strained with the Flexcell vacuum compression system. Sub-confluent cell constructs were wounded using sterile 1ml pipette tips to create an area devoid of cells. Spatial wound edge changes were monitored to determine closure rate at

Cells were isolated using percoll gradient centrifugation. Cells were seeded on elastic silicon dishes. The cells were longitudinally strained (5%, 1 Hz) for 15’ or 60’. As a control, no strain was applied. Also repetitive strain for 8 hours at three consecutive days was carried out.
